An extracellular vesicle (EV) comprises a fusion protein for therapeutic nucleic acid delivery from the extracellular vesicle which comprises a nucleic acid binding domain chosen from PUF (Pumilio and FBF Homology), Cas 6, Cas 13 and/or a nucleic acid aptamer-binding domain. The fusion protein per se, methods of loading cells with nucleic acids and pharmaceutical compositions comprising the extracellular vesicles are all disclosed. Embodiments comprise fusion proteins of FUFx2-CD63-FUFx2, PUF-CD63-PUF and Cas6-CD63-Cas6. Preferably, and among other nucleic acids, shRNA, miRNA, mRNA, gRNA, pri-miRNA, lncRNA and plasmid DNA may be delivered and more preferably the EV may further comprise a nucleic acid cargo molecule encoding a further therapeutic peptide. |
