abstract The present invention discloses a method of enriching and sequencing for a sequence of interest. A plurality of nucleic acid fragments, ligated to a first adapter, is introduced to an oligonucleotide. The oligonucleotide comprises a 3' portion complementary to a sequence of interest in the fragments, and a 5' portion comprising a second adapter sequence. The oligonucleotide is extended to give fragments comprising a sequence complementary to the first adapter sequence, a sequence complementary to the sequence of interest, and the second adapter sequence. The products are amplified using primers complementary to the adapters. The products of amplification have 3' sequence complementary to a sequence on a surface. After annealing to the surface, the products are sequenced via massively parallel sequencing.
