abstract | An improved genetic engineering platform designed to allow the interchange of promoter, polylinker, bacterial origin or replication, and bacterial antibiotic resistance gene core components within a plasmid vector using endonuclease restriction enzyme sites of eight nucleotide base pairs in length positioned between the core components of the plasmid is disclosed. The plasmid comprises a minimum of four unique endonuclease restriction sites each eight nucleotides in length, one between each of the promoter, polylinker or multiple cloning site, the bacterial origin of replication and the antibiotic resistance or selection gene. The eight-nucleotide endonuclease restriction sites may comprise restriction sites for the restriction enzymes AsiS1, Not1, Pac1, and Asc1. Viral vectors comprising three unique endonuclease restriction sites identical in sequence and relative order to the unique eight-nucleotide endonuclease restriction sites of the aforementioned vector that flank the promoter and polylinker are also claimed. |
