abstract A detection system for a polymerase chain reaction (PCR) process using fluorescence energy transfer (FRET) labelling comprising at least singly labelled oligonucleotides of differing melting temperatures (Tm) that can hybridise to each other in solution to form a fluorescent quenched pair but upon introduction of a complementary target sequence to at least one of the oligonucleotides generates a measurable signal, wherein the Tm of at least one oligonucleotide is below the annealing temperature (Ta) of the PCR process. The PCR process may be SNP genotyping or allelic discrimination. The reaction may be monitored by the 5' nuclease assay or by hybridisation.
