abstract A laser microscope with linear scanning illuminates a sample by a first and a second illumination light, LQ1, LQ2, preferably pulsed laser beams, the first effecting excitation of the sample, the second being diffracted on a periodic structure. Preferably first and second lights LQ1, LQ2, move together over the sample and are spatially superimposed in the sample, a return being made to the excited state in the sample in the regions of the second illumination light pattern. Preferably the second illumination light, which could be structured as an interferogram, comprises a periodic structure in a lateral beam direction and in an axial beam direction, the diffraction orders may be generated on a grid G and a Talbot grid effect is preferably produced. Preferably excitation and depopulation are alternately generated in the sample, achieved where both pulsed lasers are synchronised. Preferably optical resolution can be adjusted by varying the frequency of the periodic structure.
