An assay for ADP is described in which the ADP is detected by means of an ADP binding protein. A direct assay may be used in which labelled ADP, generated from a labelled precursor, is detected when the ADP is bound to the ADP binding protein. Alternatively, an indirect competition assay in which a labelled competitor of ADP is used. If ATP is present in the sample the assay may include a step of modifying the ATP, by using methionine adenosyl transferase (MAT), such that it cannot bind to the ADP binding protein. The ADP binding protein may be an antibody raised to the ADP analogue 3' dephosphocoenzyme A (DePhosCoA) or HSP90, ADP dependent glucokinase or platelet membrane ADP binding protein. Further inventions comprise production of an antibody against DePhosCoA, an affinity column comprising DePhosCoA and using MAT to degrade ATP in an ADP assay sample. |
