A chimeric polypeptide having a first and a second modified growth hormone receptor binding domain from growth hormone wherein the modification may be a deletion, substitution or addition of at least one amino acid residue and the said binding domains are joined in tandem. The binding domain may be modified in one of either site 1 or site 2 or at both sites 1 and 2. Specific modifications of said sites are disclosed as are linkers, polynucleotides encoding said polypeptides, vectors, cells expressing said polypeptide and methods of expressing said polypeptides. Pharmaceutical compositions comprising said polypeptides and their uses in treating diseases such as giantism, acromegaly, cancer, diabetic retinopathy, nephropathy or complications are claimed. The polypeptide may have a plurality of modified binding domains, especially those modified at site 2. |
