abstract | A process for the preparation of a 5-substituted-1-4-fluorophenyl)-1,3-dihydro-isobenzofuran, an intermediate for the manufacture of Citalopram, from a corresponding 5-substituted phthalide comprises carrying out a Grignard reaction on the 5-substituted phthalide in a co-solvent system comprising (i) a 4-fluorophenyl magnesium halide in an ether solvent and (ii) a suitable organic co-solvent to the ether solvent for the 5-substituted phthalide. A process for the preparation of 4-(bromo or cyano)-2-hydroxymethylphenyl-1-(4'-fluorophenyl)methanol from a corresponding 5-substituted phthalide comprises carrying out a Grignard reaction as indicated above to form a hydroxy ketone and reducing the hydroxy ketone with sodium borohydride. The cosolvent is preferably an aliphatic or aromatic chlorinated solvent or an aromatic hydrocarbon. The products are obtained in high yield and purity; and are intermediates in the preparation of Citalopram. |
