Decellularisation of tissue implant material |
abstract |
A method is described for the extraction of cellular components from body tissues and structures which are to be used as implant material. The method uses an anionic detergent, such as sodium dodecyl sulphate (SDS) or sodium deoxycholate, at a concentration which maintains the histoarchtitecture of the extracellular matrix (ECM) in conjunction with protease inhibitors, such as ethylene diamine tetraacetic acid (EDTA) and Aprotinin. Nucleic acids are removed by the use of DNase and RNase. The decellularised tissue can be stored in a cryoprotectant such as liquid nitrogen. Decellularisation improves biocompatibility and reduces immunological reactions against tissue grafts. |
