abstract | DNA constructs comprising a first promoter and a second promoter which are in opposite orientation to each other, an inter-promoter located downstream of the 3' end of the first promoter and downstream of the 3' end of the second promoter and containing at least one cloning site. Additionally the construct comprises a first transcription terminator operably linked to the first promoter located downstream of 3' end of the first promoter and downstream of the cloning site. In a preferred embodiment there is a further transcription terminator operably linked to the second promoter which is located downstream of the 3' end of the second promoter. The inter-promoter region may comprise at least two restriction sites, an <I>att</I>R1 and <I>att</I>R2 recombination sequence, and a <I>ccdB</I> sequence. The vector can be used for the production of double stranded RNA for RNA inhibition. |
