abstract |
A method of identifying a mutation which suppresses the phenotype of a nematode worm which is characteristic of the phenotype generated by exposure of a wild-type worm to a chemical comprises (a) subjecting worms to random mutagenesis; (b) dispensing one mutagenised F1 worm into each well of a multi-well assay plate; (c) allowing the worms to generate F2 offspring; (d) contacting the F2 worms with said chemical; (e) identifying worms exhibiting no or only minor changes in phenotypic, physiological, biochemical or behavioural characteristics using non-visual means; and (f) isolating from the identified worms a gene which is mutated to identify said suppressor mutation. Also claimed are methods for identifying genes that modulate the activity of a selected compound which has a defined effect on a nematode and for identifying chemicals which modulate the effect which a first compound has on the phenotype etc. of a nematode. |
