abstract | A polypeptide, having SBE ( alpha -1,4-glucan: alpha -1,4-glucan-6-glycosyl transferase; glucan branching enzyme; E.C.2.4.1.18) activity, comprises an effective portion of the amino acid sequence of Figure 4 or Figure 13 (not shown), preferably having the sequence NSKH at ca . position 697. Starch may be modified in vitro with the polypeptide. A nucleic acid sequence, obtainable from cassava and preferably as a replicable construct, encoding the polypeptide is disclosed. The sequence, operably linked in the sense or anti-sense orientation to a promoter, may be used to modify a plant host cell (preferably cassava, banana, potato, pea, tomato, maize, wheat, barley, oat, sweet potato or rice). One or more further sequences may be included, which, on transcription and/or translation, interfere with the expression of the homologous gene(s) of the host cell, preferably those involved in starch biosynthesis. |
