abstract | A process for the transformation of Burkholderia spp . (especially B.pseudomallei ) comprises: (i) culture of the bacterial cells in the presence of an antibiotic at a level at which said cells remain viable; (ii) subjecting said cultured cells to electroporation in the presence of a transforming plasmid. The antibiotic may be kanamycin, preferably in a concentration of 10 to 150 micrograms per ml. of culture medium. Electroporation may be effected using an applied voltage of from 1.4 to 1.8 kV (field strength of from 7 to 9 kV per cm.). The process may be used either to effect transposon mutagenesis or to inactivate genes by insertional mutation. The transformed bacterium may be incorporated into a vaccine, preferably in admixture with a pharmaceutically-active carrier. |
