abstract There is disclosed a method for detecting or quantifying organisms such as Erwinia carotovora atroseptica subspecies (Eca). Wells of a microtitre plate are coated with antibodies which bind Eca antigens. Samples of vegetable material are added to the well along with a a medium e.g. D-PEM (double polypectate) which selectively amplifies the bacterial cells in situ. Following a suitable period for cultivation, a labelled antibody is added to the wells. By parallel testing wells which have been enriched or non-enriched with D-PEM, the presence or quantity of live Eca cells capable of causing potato rot and 20 similar diseases can be established. The multiplication step increases the sensitivity of the test such that 100 CFU/ml can be detected. Also disclosed is the monoclonal antibody 4G4 which is Eca-specific.
