abstract | Compounds (I> <IMAGE> where R is a carboxyl group or a derivative thereof, D is C2-C18-alkylene chain; one of R1 and R2 a radical Z (which is hydrogen, deuterium, tritium, C1-C12 alkyl, C2-C4alkenyl, C2-C4alkynyl or phenyl) or a -X-C1-C6-alkyl group where X is -O- or -S-, and the other is hydrogen, deuterium, tritium, halogen, hydroxy, -X-C1-C6alkyl, with X as defined above, or a R7-COO- or R7-SO2-O- group in which R7 is C1-C6alkyl, C6-C10aryl or C7-C12arylalkyl, R1, R2 and the carbon atom to which they are bound form a <IMAGE> group, with X as defined above, a <IMAGE> group in which R8 is hydrogen, C1-C8alkyl or aryl, or a <IMAGE> group in which X1 and X2, whether the same or different, are oxygen or sulfur and A1 is C2-C18alkylene, unsubstituted or substituted with halogen; R3 is hydrogen or C1-C6alkyl; and q, R4, Y, R5, R6, E and G are various groups and the symbol ----- is a single or double bond with various provisos; and the corresponding lactones of compounds wherein R is COOH and one of R1 and R2 is hydroxy; and the usual salts and isomers thereof. These compounds are substitutes for natural prostaglandins with various advantages thereover in terms of slower metabolization and more selective therapeutic action. They are prepared by introducing the chain containing R, D, R1, R2, and R3 in place of a hydroxy group initially occupying the chain attachment point. A process is disclosed for preparing compounds (I) where one of R1 and R2 is a radical Z and the other is hydrogen deuterium or tritium by reducing the corresponding compound where R1 and/or R2 is other than Z. <IMAGE> |
