abstract | Rhodamine compounds and their uses for detecting coagulase-producing bacterial strains Rhodamine bis(tripeptide) compounds of formula I: wherein X is C(O) or S(O)1-2; Y is O, S, NH or CH2; R1a and R1b are H or C1-6alkyl, where the alkyl group may be substituted with halo; R2a and R2b are H, C(O)Ra, C(O)ORb, L-aryl or a G group; R3a, R3b, R4a and R4b are H, NO2, OH, C(O)Rc or optionally halo-substituted C1-6alkyl; m is 0-3; n is 0-4; each R5 is halo or other substituents; or two or more R5 groups may be joined together with the carbon atoms to which they are attached a 5- or 6-membered aryl or cycloalkyl group; each R6 is NHRf, NRgRh, C(O)Rh, ORi, or optionally halo substituted aryl or C1-6alkyl; Ra-Ri are optionally halo-substituted C1-6alkyl; L is a direct bond or CH2; G is any amino acid which may be further substituted; may be useful for the detection of coagulase-producing bacterial strains, particularly Staphylococcus aureus. The preferred compound is rhodamine coupled to the N-Boc protected tripeptide Val-Pro-Arg. The compounds are useful in detecting methicillin resistant Staphylococcus aureus (MRSA) and MSSA. |
