abstract 1530572 Cephalosporin derivatives RHONE POULENC INDUSTRIES 17 Jan 1977 [19 Jan 1976] 01778/77 Heading C2C Novel cephalosporin derivatives of the general Formula I wherein R 1 is acetoxy, (5-methyl-1,3,4-thiadiazol-2-yl)-thio, (1-methyl-tetrazol-5-yl)-thio or [1-(2-hydroxyethyl)-tetrazol-5-yl]-thio and R 2 is carboxyl; or R 1 is pyridinio and R 2 is the carboxylato ion; and, when R 2 is carboxyl, their metal salts and addition salts with nitrogencontaining bases are prepared (a) by reacting (1,3-dithiin-5-yl)-acetic acid or a reactive derivative thereof with a cephalosporin of the general Formula III or, when R 2 is carboxyl, a derivative thereof in which the carboxyl group is protected, followed if necessary by removal of the protecting group; or (b) when R 1 is other than acetoxy, by reacting the corresponding compound in which R 1 is acetoxy with 5-methyl-2-thioxo-1,3,4- #4-thiadiazoline, 1-methyl-5-thioxo-tetrazoline, 1-(2-hydroxyethyl)-5-thioxo-A2-tetrazoline or pyridine; followed optionally by salification of the product. The preparation of (1,3-dithiin-5-yl)-acetic acid and the acid chloride thereof is described. Pharmaceutical compositions having antibacterial activity comprise, as active ingredient, a cephalosporin derivative (I) or pharmaceutically acceptable salt (as defined above) thereof, in association with a compatible pharmaceutically acceptable diluent or adjuvant.
