titleOfInvention 18-methyl-19-nor-20-keto-pregnanes and process for their manufacture
abstract 1512258 18 - Methyl - 19 - nor - 20 - ketopregnanes SCHERING AG 4 Aug 1975 [5 Aug 1974] 32477/75 Heading C2U Novel steroids of the formula (wherein R 1 and R 2 together are oxo, or one of them is H and the other OH, R 3 is oxo or H 2 and 5-H is α or #) are prepared by treating steroids of the formula which contain an optional #<SP>4</SP>-double bond, with an alkali or alkaline earth metal in liquid ammonia and, if desired, re-oxidizing any keto group that is reduced by the treatment, or by hydrogenating steroids of the formula which contain a #<SP>4</SP>- and/or a #<SP>16</SP>-double bond, and, if desired, selectively reducing a resulting 3-ketone to a 3-ol. In one example a resulting 3#-ol is converted to 3α-formyloxy-18-methyl- 19 - nor - 5α - pregnan - 20 - one and this is then hydrolysed to the corresponding 3α-ol. 17# - Acetoxy - 18 - methyl - 19 - nor - 5#,17α- pregnane - 3,20 - dione and the corresponding 5α - compound are prepared from 17α - ethynyl - 17# - acetoxy - 18 - methyl - #<SP>4</SP>- estren - 3 - one by converting this to 17#- acetoxy - 18 - methyl - 19 - nor - 17α - #<SP>4</SP> - pregnen - 3,20 - dione and catalytically hydrogenating this. 18 - Methyl - 19 - nor - #<SP>4'16</SP>- pregnadiene - 3,20 - dione is prepared from 17- ethynyl - 18 - methyl - #<SP>4'16</SP> - cotradien - 3 - one. The novel steroids have a central-depressive anaesthetic-narcotic action and anticonvulsive activity, and they may be made up into pharmaceutical compositions with suitable carriers.
