titleOfInvention Orthoester polymers
abstract 1502082 Sustained release compositions ALZA CORP 8 Jan 1976 [28 Jan 1975] 00683/76 Heading A5B [Also in Division C3] Sustained release compositions contain active ingredients and polymers of formula where R<SP>1</SP> is a di- or poly-valent organic radical R<SP>2</SP> and R<SP>3</SP> are monovalent radicals or are linked together (as shown by the dotted line) to form a heterocyclic ring and n is at least 10. The radical R<SP>1</SP> may be alkylene, alkenylene, oxaalkylene, cycloalkylene, cycloalkenylene, cycloalkanedi - aliphatic hydrocarbylene, phenylene or phenyldi - aliphatic hydrocarbylene and may be substituted by alkyl, alkoxy or alkenyl groups; R<SP>2</SP> and R<SP>3</SP> when not directly linked together may be alkyl, alkenyl or alkenyloxy groups or -OR<SP>1</SP>O-, at least one being alkenyloxy or -OR<SP>1</SP>O-, R<SP>2</SP> and R<SP>3</SP> when linked together form with the carbon atom of the polymer chain a hetero-mono-or polycyclic group with I or 2 oxygen heteroatom linkages to the carbon atom of the polymer chain the remaining annular atoms being carbon atoms optionally substituted by alkyl, alkoxy or alkenyl groups. The polymers may be homopolymers or random or block copolymers. The polymers are bioerodible and the active ingredient e.g. a drug, is released slowly as the polymer erodes e.g. in or on a living body. In Examples there are described devices for application to the skin, an ointment, solid implants and ocular inserts.
