abstract 1490015 Heterocyclic amidines UPJOHN CO 5 Feb 1975 [11 Feb 1974] 4846/75 Heading C2C The invention comprises compounds of formula wherein R 1 and R 2 are each H, C 1-8 alkyl, C 5-8 cycloalkyl, phenyl, mono- or di-substituted phenyl, phenyl (C 1-3 ) alkyl or mono- or disubstituted phenyl (C 1-3 ) alkyl each substituent being selected from C 1-3 alkyl, C 1-3 alkoxy, halogen and CF 3 and at least one of R 1 and R 2 being other than H and Z is morpholino, thiomorpholino, pyrrolidino, piperidino, perhydroazepino, piperazino or N-(C 1-3 ) alkylpiperazino and acid addition salts thereof, e.g. hydrochloride maleate and nitrate. The compounds may be prepared by reacting the corresponding carbodiimide (R 1 N = C = NR 2 ) and heterocyclic amine ZH. The carbodiimide may in turn be prepared from the corresponding urea or thiourea (R 1 NHCXNHR 2 ) and the thiourea may be prepared from the corresponding amine R 1 NH 2 and isothiocyanate (R 2 NCS). Alternatively the compounds of the invention may be prepared by treating the corresponding heterocyclic amide R 1 NHCOZ with phosgene to provide the carboximidoyl chloride and reacting this with the amine R 2 NH 2 . Pharmaceutical compositions containing the compounds of the invention are useful to treat arrythmia and as diuretics.
