abstract 1480003 Tetracyclic Diels-Alder adducts MERCK & CO Inc 1 Oct 1975 [4 Oct 1974 9 May 1975] 40160/75 Heading C2C The invention comprises compounds of formula and their non-toxic pharmaceutically acceptable salts, esters and amides, wherein R<SP>1</SP>, R<SP>2</SP>, R<SP>3</SP> and R<SP>4</SP> are each H, halogen, alkyl, aminoalkyl, carboxyalkyl, aralkyl, cycloalkyl, haloalkyl, acyloxy, alkenyl or dialkylaminoalkyl; X is CH 2 or CH:CH; and R is propargyl, amino- or acylamino - phenyl (optionally further substituted by halogen C 1-6 alkyl, alkoxy, aminoalkyl, carboxyalkyl, carbalkoxy, carbamoyl or haloalkyl), 2 - amino- or -acylamino - 5- pyrimidinyl, 5- or 6 - (2 - oxo)benzoxazolinyl, 2-thiazolyl or 5-(2-oxo)indolinyl. In examples, these compounds are prepared by (a) Diels- Alder reaction, (b) reaction of the analogous cyclic anhydride with RNH 2 (or a suitable derivative thereof), or (c) (when R=aminophenyl) by hydrolysing the acetaminophenyl compound. Starting materials otherwise prepared have the formul4 2,4-X,YC 6 H 3 NHCOBu, where X is H or Cl and Y is NH 2 or NO 2 . Therapeutic compositions having minor tranquillizing and anticonvulsant activity comprise compounds of the above formula, and may be administered orally, parenterally or rectally.
