abstract 1479994 Determination of total cholesterol ABBOTT LABORATORIES 25 Feb 1975 [4 March 1974] 7904/75 Heading G1B Total cholesterol is enzymatically determined in a serum sample using a single reagent comprising a substantially anhydrous mixture of cholesterol esterase, cholesterol oxidase, horse-radish peroxidase (HRP), a hydrogen donor oxidisable by hyrdogen peroxide to produce a colour, and cholic acid, a conjugate thereof or a salt of either, the cholesterol esters in the serum sample being hydrolyzed to free cholesterol which is then oxidized to # 4- cholestenone and hydrogen peroxide, the hydrogen peroxide in the presence of HRP oxidizing the hydrogen donor which may be o-diamisidine or 4-aminoantipyrine + phenol to form a colour which is measured. The assay of cholesterol oxidase by conversion of cholesterol to cholest-4-en-3-one and measurement of the latter at 240 nm is disclosed. Hydrogen donors other than those mentioned above are 4', 4', 4" methylidynetris (N,N-dimethylaniline), 3-methyl-2-benzo-thiazolinone hydrazone/o-toluidine, 3-methyl-2-benzothiazolinone hydrazone/ N,N-dimethyl aniline, 2, 2' -azino-di-(3-ethylbenzothiazolin sulfonate-6) diammonium salt.
