abstract 1472574 Isolating snake venom enzyme LABORATORI BIOCHIMICI FARGALPHARMASINT SpA 5 June 1974 [3 Sept 1973] 24920/74 Heading C3H [Also in Division A5] A process for extracting from snake venom, e.g. from Ancistrodon rhodostoma the enzyme component having in vitro coagulant activity and in vivo anti-coagulant activity comprises chromatographing the venom on a column comprising a support, insoluble in aqueous solutions and having bound thereto a substrate or inhibitor for the enzyme by eluting first with an aqueous buffer solution of pH 6-7 and a molarity of 0À05% until more than 95% of the protein components and in vitro coagulant fractions, different from the desired enzyme has been obtained and secondly with a buffer of pH greater than 9 and a molarity greater than or equal to 0À3 M until the enzyme with in vivo anti-coagulant activity together with less than 5% of protein components is obtained.
