| abstract | 1461444 Pregnane 18-azides; amines; iodides; conanines CHOAYSA 8Feb 1974 [8 Feb 1973] 05912/74 Heading C2U The invention comprises 18-azido steroids of the pregnane series having at the 20-position an oxo group or a free, esterified or etherified hydroxyl group; and their preparation from 18- iodo steroids of the pregnane series having at the 20-position a protected hydroxyl group by reaction with an alkali metal azide, followed as required by hydrolysis of the 20-protected group to a free hydroxy group and oxidation of the latter. 18 - Iodo - 20# - acetoxy - 5α - pregnane, 18 - iodo - 3#,20# - diacetoxy - 5α - pregnane and 18 - iodo - 3# - acetoxy - 20 - (2 - tetrahydropyranyloxy) - 5α - pregnane are prepared from the corresponding 20-ols by reaction with Ac 2 O or dihydropyran. 5α - Conanin - 3# - ol (XIV) is prepared from 18 - azido - 3#,20# - diacetoxy - 5α - pregnane (XI) by the sequence XI#18 - amino - 3#,20#- dihydroxy - 5α - pregnane#5α - norcon - N(20)- enin - 3 - one (XIII)#5α - norcon - N(20)- enin - 3# - ol(?)#XIV, the last two steps being by the actions of NaBH 4 and HCHO/ HCO 2 H respectively. 3,3 - Dimethoxy - 5α - conanine is prepared from XIII by ketalization to give 3,3-dimethoxy - 5α - norcon - N(20) - enine and reaction of the latter with HCHO/HCO 2 H. Hypotensive parenteral compositions comprise a compound of the invention in alcoholic solution. |
