abstract 1451835 Enzymatic hydrolysis of RNA KYOWA HAKKO KOGYO CO Ltd 13 Feb 1974 [14 Feb 1973 24 March 1973] 6571/74 Heading C2P Nucleoside 2<SP>1</SP>,3<SP>1</SP>-cyclic phosphates and nucleoside 3<SP>1</SP>,5<SP>1</SP>-cyclic phosphates are obtained by enzymatic hydrolysis of ribonucleic acid by reacting it with intact cells of a microorganism having a ribonuclease activity in an aqueous reaction medium in the presence of a chelating agent. The reaction medium may also contain a source of phosphate ion, the amount of phosphate ion influencing the product formed. In the presence of 0 to 0À15 M phosphate ion the 2<SP>1</SP>,3<SP>1</SP>-cyclic phosphate is selectively produced and in the presence of at least 0À17 M phosphate ion the 3<SP>1</SP>,5<SP>1</SP>-cyclic phosphate is selectively produced. The microorganism may be a bacterium of one of the genera Escherichia, Salmonella, Aerobacter or Pseudomonas, such as Escherochia coli, Aerobacter aerogenes, Salmonella typhosa or Pseudomonas aeruginosa or a yeast of one of the genera Hansenula, Kloeckera or Torulopsis, such as Hansenula anomala, Kloeckera apiculata or Torulopsis sphaerica. The chelating agent may be ethylenediaminetetraacetic acid, cyclohexanediaminetetraacetic acid or hydroxyethylenediaminetriacetic acid. The pH of the medium is preferably maintained at 4 to 9, more preferably about 5 for the production of the 2<SP>1</SP>,3<SP>1</SP>-cyclic phosphate and about 8 for the production of the 3<SP>1</SP>,5<SP>1</SP>-cyclic phosphate.
