abstract | 1422518 Prostaglandins AYERST MCKENNA & HARRISON Ltd 23 March 1973 [27 March 1972] 14128/73 Heading C2C Compounds I in which (a) is CH 2 CH 2 , cis CH = CH or C # C, p is 2-4, (b) is trans CH=CH, (c) is either (CH 2 ) q where q is 1-6, or cis CH 2 CH = CH(CH 2 ) r where r is 0-3, R is H or C 1-6 alkyl and R<SP>2</SP> is H, C 1-6 alkyl or CH 2 OR<SP>3</SP> where R<SP>3</SP> is H or C 2-5 alkanoyl, provided that R<SP>1</SP> is the same as R<SP>3</SP>, are prepared by subjecting a compound II in which R<SP>2</SP> is H, C 1-6 alkyl or CH 2 O-(C 2-5 alkanoyl), to a base-catalysed condensation with a compound III to give a compound of Formula IVa or IVb in which R<SP>7</SP>=H or C 1-6 alkyl, compound IVa being obtained when R<SP>2</SP> in compound II is H or C 1-6 alkyl and compound IVb being obtained when R<SP>2</SP> in compound II is alkanoyloxymethyl; when R<SP>5</SP> is not H, subjecting the compound IVa or IVb to treatment with an acid to remove the protecting radical, and then treating the resulting compound with a base under aqueous conditions to give a compound I in which R and R<SP>1</SP> are H and R<SP>2</SP> is H, C 1-6 alkyl or CH 2 OH, if desired, estifying this compound to give a compound I in which R is C 1-6 alkyl, and, if desired, acylating a compound I in which R is H or alkyl to give the corresponding compound I in which R is H or alkyl, R<SP>1</SP> is alkanoyl and R<SP>2</SP> is H, alkyl or alkanoyloxymethyl. The compounds trans-7-[2-(3-hydroxy-1-octenyl) - 3 - hydroxymethyl - 5 - oxo - cyclopentyl]heptanoic acid and its methyl ester, and trans - 7 - [2 - (3 - hydroxy - 1 - octenyl) - 5- oxocyclopentyl]-5-heptynoic acid are claimed per se. |
