abstract 1421488 Making carrier-bonded polypeptides by irradiation; modified acrylamide copolymers ROHM GmbH 21 Dec 1973 [23 Dec 1972] 59522/73 Headings C3H and C3P A process for the preparation of carrierbonded polypeptides, for example an enzyme, antibody, hormone or enzyme inhibitor comprises irradiating in an essentially oxygen-free atmosphere an aqueous solution of a polypeptide containing a non-peptidic macromolecular reactive carrier, the said solution being irradiated with UV or visible light in the presence of an organic peroxide. The carrier may be a water soluble, water-swellable or water insoluble macromolecular compound preferably containing carboxylic amide, ester, lactone stemiacetal and/or cyclic ether groups and may be a polysaccharide, e.g. a dextran gel, agarose gel or cellulose, vinyl pyrrolidone or may comprise units of methacrylamide, acrylic or methacrylic acid or water-soluble salts thereof, a hydroxyalkyl ester of acrylic or methacrylic acid, 1,2 or 1,4-butanediol acrylate, or methacrylate, an aminoalkyl ester or aminoalkyl amide of acrylic or methacrylic acid or a quaternization product thereof, or units of divinyl benzene, glycol diacrylate or dimethacrylate, triallyl cyanurate, and may comprise side alkenyl and/or alkylphenyl groups. The irradiation may be performed in the presence, as sensitizer, of propiophenone, xanthene, acetophenone, triacetylbenzene, isobutyrophenone, diphenylpropanone benzaldehyde, triphenylmethyl ketone, carbazole, diphenylene oxide, triphenylamine, dibenzothiophene, o-dibenzoylbenzene, benzophenone, dichlorobenzophenone, diacetylbenzene or flucrene. In Example 8, an acrylamide/2-hydroxyethyl methacrylate copolymer is modified by reaction with p-toluene sulphonyl chloride. In Example 9, a similar copolymer is modified by reaction with 4-methylphenylacetic acid chloride.
