abstract 1420885 Administering drugs to the eye; pharmaceutical preparations ALZA CORP 28 Jan 1974 03837/74 Headings A5B and A5R A drug is dispensed to an eye at a predetermined rate for a prolonged period of time by a sealed container 10 insertable in a cul-de-sac of the eye and having first and third walls 11, 14, each insoluble in tear fluid and at least one being permeable to the drug by diffusion, separated by an annular second wall 12, with a drug mixture 13 comprising a drug and carrier in the interior space 15 formed by the walls 11, 12, 14. The permeable wall(s) may comprise a homogeneous imperforate natural or synthetic polymer, of which examples are given, e.g. natural rubber, silicone rubber, plasticised nylon, polymethylmethacrylate, or ethylene-vinylacetate copolymer. Alternatively the permeable walls may comprise microporous material with pore sizes from 10Š to 100 microns, of which examples are given, e.g. celluloses, cellulose acetate propionate or polycarbonates, filled with a diffusive medium in which the drug has a limited solubility, of which several examples are given, e.g. water, glycerin or propylene glycol. The second wall 12 comprises a cold or hot setting adhesive polymeric material (setting temperature 5‹C to 250‹C) heat sealed to the walls 11, 14, examples of materials being given, e.g. polyvinylacetate or polyamides, and may include a dye for identification purposes. The drug may comprise antibiotics e.g. tetracycline, antibacterials e.g. sulfonamides, antivirals e.g. idoxuridine, anti-allergenics e.g. antazoline, antiinflammatories e.g. hydrocortisone, decongestants e.g. phenylephrine, miotics and anticholinesterases e.g. pilocarpine, mydriatics e.g. atropine sulfate, or sympathomimetics e.g. epinephrine, suspended in a solid or semi-solid carrier, e.g. gelatin, starches, pectin &c. The device may be of various shapes, e.g. elliptical, bean-shaped, banana-shaped, circular, rectangular, trapezoidal or doughnut shaped, for retention in the eye.
