abstract | 1385209 Diagnostic agents BEHRING- WERKE AG 12 May 1972 [12 May 1971] 22380/72 Heading A5B [Also in Divisions C3 and G1] A protein or peptide-polystyrene complex, which may be used as a diagnostic agent, comprises a protein or peptide which is linked by means of an aromatic diazonium compound to a homopolymer of styrene or a copolymer of styrene, having more than 50% of styrene units, with such monomers as acrylic acid or methacrylic acid (Example 8). A polystyrene suspension may first be reacted with the diazonium compound in the presence of glycerol as stabiliser and the protein or peptide then reacted with the diazotised polystyrene. Theproduct is stabilized by addition of 20% human serum albumin solution and homogenised ultrasonically. Diazonium compounds mentioned in the Examples: diazotised-(1) p-aminophenyl-sulphone - ethoxy-sulphonic acid, (2) 7- ethoxy-3, 9-diamino-acridine lactate, (3) benzidine, disanisidine, 4, 4'-diamino-diphenyl-sulphone, 4-aminophenyl-sulphone-ethoxy-sulphonic acid, 2, 5-dichloro-4-nitroaniline, aniline, naphthylamine and 7-ethoxy-3, 9-diamino-acridine, (4 to 8) benzidine. Other diazonium compounds are particularly mentioned. The protein or peptide may be an albumin, globulin, proteshormone, or a viral, bacterial, protozoan or helminthic protein. The following diagnostic agents are specified in the Examples: (1) human choriogonadotropin, crystallised zinc insulin, anti-immunoglobin M and anti-hepatitis - associated antigen (HAA), (2) anti-HAA, (3) anti- HAA, zinc insulin, anti IgE or IgA, anti-alphafoetoprotein, anti-pregnancy specific protein, anti-human placenta lactogen, anti-fibrinogensplit D or E, anti-myoglobin, anti-haptoglobin, anti-C-reactive protein, anti-alpha-anti-trypsin and anti-transaminase, (4) human choriogonadotropin, rubella virus protein and alpha-hemolysin obtained from Staphylococci, (5) Trypanosoma reagent, (6) Schistosoma reagent, (7) protein factor from Entamoeba histolytica and (8) anti-HAA. The complex may be admixed with a pharmaceutically suitable diluent to form the diagnostic reagent. |
