1374454 L - tryptophan and derivatives AJINOMOTO CO Inc 21 Dec 1971 [22 Dec 1970] 59436/71 Heading C2C [Also in Division C3] L-tryptophans of the formula where R is hydrogen, hydroxy, amino or methyl are prepared by reacting the corresponding indole with ammonium ions and at least one of pyruvic acid, oxaloacetic acid, malic acid, fumaric acid, glyoxylic acid, lactic acid and salts and other derivatives thereof, in an aqueous solution in the presence of tryptophanase or a source thereof. Vitamin B 6 , inosine and/or reducing agents may be present. The enzyme source may be the enzyme itself, a broth in which a microorganism capable of producing the enzyme is cultivated, a cell-free filtrate thereof, an aqueous suspension of the cells or of ground cells, or a filtrate thereof. Suitable microorganisms belong to the genera Escherichia, Aerobacter, Erwinia, Pseudomonas, Xanthpmonas, Proteus, Citrobacter, Paracolobactrum, Salmonella, Clostridium and Kluyvera. In examples in which inosine is present in the production of L-tryptophan, a L-tryptophaninosine complex is precipitated. |
