titleOfInvention Process for producing 7-aminodesacetoxy-cephalosporanic acid
abstract 1369970 Producing 7-amino-desacetoxycephalosporanic acid TOYO JOZO KK 18 June 1973 [17 June 1972] 28747/73 Heading C2A [Also in Division C3] 7-Amino-desacetoxycephalosporanic acid or an alkali metal salt thereof is produced by N- deacylating 7-phenoxy-acetamido- or 7-phenylacetamido - 3 - methyl - #<SP>3</SP> - cephem - 4 - carboxylic acid or an alkali metal salt thereof by treatment in aqueous solution with a deacylating enzyme from Arthrobacter simplex ATCC 15799, Kluyvera citrophila ATCC 21285, Proteus rettgeri ATCC 9250 or Micrococcus sp. NRRL B-2753. The enzyme may be in the form of a cultured whole broth, the filtrate thereof, the separated cells or a separated enzyme preparation consisting of the enzyme adsorbed on a suitable carrier (e.g. diatomaceous earth, an ionexchange resin or an ion-exchanging cellulose derivative). The deacylation is suitably effected at pH 7 to 8 and 20‹ to 45‹ C., by incubating the aqueous acylamino-cephalosporin solution with the enzyme preparation or, in the case of the carrier-bound enzyme, by passing the solution through a column thereof. The product may be isolated by precipitation with acetone.
