abstract | 1346862 Immunological reagent F HOFFMANN-LA ROCHE & CO AG 31 Jan 1972 [1 Feb 1971] 4416/72 Heading B1X [Also in Division C3] Water-insoluble diagnostic reagents having a specific gravity close to that of water comprise discrete particles of a serologically inert latex polymer (selected from those listed below) having condensed thereto through an amide linkage a known serologically determinant material. They are made by contacting the polymer with the determinant material at 5-40‹C in the presence of a carbodiimide. The product may be made up into a test kit, which may be a two-part kit containing in the first container a product of the invention and in the second container anti-serum for the determinant material. The determinant material may be selected from human and animal antibodies, serum components, toxins, bacterial and viral components, hormones, enzymes alkaloids, cellular and tissue extracts, and low M.W. substances such as insulin, angiotension and urokinase. Those used in specific Examples are human chorionic gonadotropin, human albumin and denatured gamma globulin. The polymer latex is one having an average particle size in the range 0.01-0.9 microns and selected from carboxylated styrene/butadiene copolymers, carboxylated polystyrene optionally containing amino groups, acrylic or methacrylic acid polymers, acrylonitrile polymers, ABS, polyvinyl pyridines, and acrylate/vinyl acetate or chloride copolymers. Those used in specific Examples are carboxylated styrene/butadiene latex or acrylonitrile latex. |
