abstract 1345340 13 - Polycarbonalkyl - 16 - methylgona-1,3,5(10)-trienes AMERICAN HOME PRODUCTS CORP 23 Sept 1971 [23 Sept 1970] 21061/73 Divided out of 1345339 Heading C2U Novel steroids of the formula (wherein R 1 is C 1-6 alkyl or a C 3-7 cycloaliphatic radical, R 2 is C 2-18 alkyl and R 3 is C 1-6 alkyl or C 2-6 alkynyl) are prepared by reductive alkylation or alkynylation of steroids of the formula to give 16#-methyl-17α-alkyl-17#-ols or 16(α or #) - methyl - 17α - alkynyl - 17# - ols and, when required, hydrogenation of the latter to give 16(α or #) - methyl - 17α - alkyl - 17# - ols. In the alkynylation process epimerization of the 16-methyl substituent may occur, depending on the reagent used. Thus, use of MgBrC#CBrMg results in a 16#-methyl-17α-ethyryl-17#-ol as the major product while use of lithium acetylideethylenediamine complex results in a 16α- methyl-17α-ethynyl-17#-ol. Steroids of the second general formula above are prepared by catalytic hydrogenation of steroids of the formula which are in turn prepared by subjecting corresponding 16-unsubstituted steroids to a Mannich reaction to give steroids of the formula and treating these with a base. In examples illustrating the above processes, the starting materials, intermediates and products are in racemic form. The novel steroids are stated to possess hormonal activity as estrogens and/or antilipaemics. They may be made up into pharmaceutical compositions with suitable carriers.
