abstract 1336443 3<SP>1</SP>,5<SP>1</SP>-Nucleotides; purine derivatives AGENCE NATIONALE DE VALORISATION DE LA RECHERCHE 7 Oct 1970 [10 Oct 1969 2 Oct 1970] 13401/73 Divided out of 1336441 Headings C2P and C2C 3<SP>1</SP>,5<SP>1</SP>-Nucleotides and salts thereof are prepared by cyclizing a 5<SP>1</SP>-nucleotide or salt thereof by reaction with dicyclohexylcarbodiimide in the presence of pyridine and dimethyl formamide. The 3<SP>1</SP>,5<SP>1</SP>-nucleotide may have the Formula I: where one of X and Y is H or alkyl and the other is alkyl and R 1 and R 2 are both H, or X and Y are both H, R 1 is H, Br, SH, OH, SCH 3 or NH 2 and R 2 is H or NH 2 . The starting material 2-aminoadenosine-5<SP>1</SP>- monophosphate is prepared (isolated as its triethylammonium salt) by reacting 2-amino-6- chloro - 9 (2<SP>1</SP>,3<SP>1</SP>,5<SP>1</SP> - triacetyl - beta - D - ribofuranosyl)purine with a saturated solution of ammonia to deacetylate it, reacting the 2- amino-6-chloro nucleoside produced with concentrated ammonia in an autoclave to produce the corresponding 2,6-diamino-nucleoside, reacting this with dimethoxy propane to produce the corresponding 2<SP>1</SP>,3<SP>1</SP>-isopropylidene derivative, and finally phosphorylating this product with phosphorus oxychloride, the isopropylidene groups being eliminated by warming.
