abstract 1326923 Deproteinising samples for analysis; determination of serum phosphate EI DU PONT DE NEMOURS & CO 28 July 1970 [19 Aug 1969] 36593/70 Heading B1X A liquid sample containing interfering charged macromolecules is analysed for a particular constituent by a process comprising forcing the sample through an ion exchange medium into a reaction chamber, the ion exchange medium being one capable of removing at least one type of charged macromolecule from the sample, and determining the constituent in the liquid in the reaction chamber. The apparatus of Fig. 1 comprises ion exchange resin column 14, reaction chambers 15, liquid handling means which may comprise pump 13, probe 12 for removing a predetermined amount of sample to be analysed from a container 11 and forcing it through column 14, for example by a following stream of diluent, means 16, 17, 18 for introducing at least reagent into the liquid in chambers 15, and means for determining a constituent in the resulting mixture, the ion-exchange resin used in the column comprising oarboxymethyl cellulose, carboxymethyldextran, sulfoethylcellulose, sulfoethyl dextran, polystyrene divinyl benzene sulfonic acid, diethyl aminoethyl cellulose, diethyl aminoethyl dextran, cellulose phosphate or ecteola cellulose. For the determination of serum phosphate, the serum sample is passed through a column of sulfoethylcellulose and polystyrene divinyl benzene sulfonic acid, using water as eluent, reagents comprising trichloroacetic acid, ascorbic acid, ammonium molybdate and arsenite-citrate are added to the eluted sample and the absorbance of the reaction mixture is measured at 600A. The process of the invention may be used with the reagent package of 1188985, when the ion exchange resin is located in the header member.
