abstract 1324159 Producing 7-amino-desacetoxycephalosporanic acid TOYO JOZO CO Ltd 14 March 1972 [31 May 1971] 11915/72 Heading C2A [Also in Division C3] 7 - Amino - desacetoxycephalosporanic acid having the formula is produced by N-deacylating a water-soluble salt of 7-phenylacetamido- or 7-phenoxyacetamido-desacetoxycephalosporanic acid with a deacylating enzyme produced by Bacillus megaterium B-400, FERM-P No. 748. The enzyme may be in the form of a culture filtrate of the micro-organism or a purified preparation derived from the filtrate, e.g. the enzyme adsorbed on a non-inactivating carrier (see Division C3). The preferred water-soluble salt of the cephalosporin is the Na or K salt, and the N-deacylation of the salt is effected either by incubating it insolution in a culture filtrate of the micro-organism or by passing its solution through a column of the enzyme adsorbed on an insoluble inert carrier, at pH 7-8 and 30-45‹ C. using a concentration of the salt of 0À1 to 20 mgs./ml. The 7-aminodesacetoxycephalosporin may be recovered by bringing the solution to its iso-electric point (pH 3.7), concentrating, cooling to precipitate the product and washing the precipitate with acetone and drying.
