abstract | 1320086 Determination of enzyme concentration BOEHRINGER MANNHEIM GmbH 7 Sept 1971 [11 Sept 1970 15 June 1971] 41657/ 71 Heading B1X The concentration of enzymes, e.g. endoenzymes, which hydrolise, e.g. polysaccharides, proteins or esters, is determined with an agent which comprises an enzyme-hydrolisable, water insoluble or insolubilised and optically or photographically evaluably marked substance dispersed in an inert carrier matrix which permits diffusion of any hydrolised fragments of the dispersed substance. The agent is supported by e.g. a glass or resin plate or a transparent or opaque foil. Application of test sample to the agent causes hydrolysis of the marked substance when hydrolysed fragments diffuse through the carrier matrix to leave an unmarked test area from which the concentration of enzyme can be found by comparison with standard results. The carrier matrix may be agar, agarose, pectin gelatine, starch, quarane, polyacrylamide, silicate gel, cellulose or filter paper. The dispersed substance may be starch, cellulose, dextran or a protein cross-linked with diepoxide and may be marked with a dyestuff, a fluorescent substance or a radioactive agent (all specified). If water soluble the substance to be dispersed can be insolubilised by cross-linking, e.g. with halohydrins, isocyanates, isthiocyanates or epoxides, or by binding with activated particles of dextran, cellulose, polystyrene or glass. The test sample may be urine (for α-amylase), Penicillium funiculosum (for crude dextranase) or a solution which contained papain. Other enzymes are mentioned. |
