abstract | 1308121 Total synthesis of dl-estradiol INSTITUT KHIMI PRIRODNYKH SOEDINENY AKADEMII NAUK SSSR 16 April 1971 9683/71 Headings C2C and C2U In the preparation of dl-estradiol by the sequence 6-methoxytetralone -# 6-hydroxytetralone -# 1 - vinyl - 1,2,3,4 - tetrahydronaphthalene - 1,6 - diol # 3 - hydroxy- 8,14 - secoestra - 1,3,5(10),9(11) - tetraene- 14,17 - dione # dl - 8,14 - bisdehydroestrone # dl - 8 - dehydroestrone # dl - 8 - dehydroestradiol # dl - estradiol, the first step is effected by boiling with HBr and acetic acid, the second step by reaction with vinyl magnesium bromide, the third by reaction with 2-methylcyclopentane-1,3-dione, the fourth by cyclization e.g. with HCl, the fifth by hydrogenation, e.g. using a Pd-CaCO 3 catalyst, the sixth by NaBH 4 , and the last by reduction with triethylsilane and trifluoroacetic acid in an organic medium. The final product is preferably isolated by extraction with benzene from the reaction mixture, concentration of the extract by evaporation, treatment with NaBH 4 / MeOH at 40-45‹ C. then with acetic acid while cooling to precipitate the product. |
