abstract 1305795 2,6 - Dialkyl - 4 - azidoaryl -1,4 - dihydropyridine - 3,5 - dicarboxylic acid esters BAYER AG 19 April 1971 [20 March 1970] 24540/71 Heading C2C Novel 2,6 - dialkyl - 4 - azidoaryl - 1,4 - dihydropyridine-3,5-dicarboxylic acid esters of the general formula wherein R is a hydrogen atom or a straight or branched chain or cyclic C 1-6 alkyl, alkenyl or alkynyl group, optionally substituted by one or two hydroxy groups or a C 1-3 alkoxy group, or a benzyl or phenethyl group, the aryl moiety of which is optionally substituted by 1-3 C 1-3 alkoxy and/or 1 or 2 C 1-3 alkyl groups and/or 1 or 2 halogen atoms selected from F, Cl and Br atoms; R<SP>1</SP> is a hydrogen atom or a straight or branched chain C 1-4 alkyl group; R<SP>11</SP> is a straight or branched chain or cyclic C 1-6 alkyl, alkenyl or alkynyl group, the carbon chain of which is optionally interrupted by 1 or 2 oxygen atoms, optionally substituted by an hydroxy group; and R<SP>111</SP> is an aryl group which is substituted by an azido group and optionally 1 or 2 C 1-4 alkyl or fluoroalkyl and/or 1 or 2 C 1-4 alkoxy groups and/or 1 or 2 halogen atoms selected from Cl and Br atoms, are prepared (a) by reaction of an aldehyde of the general formula R<SP>111</SP>CHO with (i) an ester of the general formula R<SP>1</SP>COCH 2 COOR<SP>11</SP> and ammonia or an amine of the general formula RNH 2 or a salt of such an amine or (ii) an enamine of the general formula in an organic solvent or water, at an elevated temperature; or (b) when R is not a hydrogen atom, by reduction of a corresponding quaternary pyridinium salt. Pharmaceutical compositions having coronary vessel dilating, anti-fibrillating, spasmolytic and anti-hypertensive activity comprise, as active ingredient, at least one 2,6-dialkyl-4-azidoaryl- 1,4-dihydropyridine-3,5-dicarboxylic acid ester of the first general formula above, in admixture with a solid or liquid pharmaceutically acceptable diluent or carrier, and may be administered orally or parenterally.
