titleOfInvention O-(nitroaryl)oximes of 3-keto steroids
abstract 1299650 3 - (Nitropenoxyimino) - steroids ORTHO PHARMACEUTICAL CORP 30 Nov 1970 [4 Dec 1969] 56679/70 Headings C2C and C2U The invention comprises compounds of formula wherein each Z is independently a carboncarbon single or double bond with the proviso that when Z in the 4-5-position represents a single bond then the 5-hydrogen atom is alpha-oriented; R 1 and R 2 are each H or NO 2 , at least one of R 1 and R 2 being NO 2 ; R 3 is H or Me; R 4 is H, Me, Et, ethynyl or acetoxy; R 5 is H or Me; R 6 is OH, acyloxy or acetyl with the pvosio that when R 4 is acetoxy then R 6 is acetyl; or R 4 and R 6 together represent OXO; R 7 is H or halo; and R 8 is H or halo. Preparation of compounds I is by (i) reaction the corresponding 3-oxo-compounds with an approriate O-aryl-hydroxylamine in the presence of an acid catalyst; or (ii) reaction of the corresponding 3-hydroxyimino compounds with an appropriate aryl halide in the presence of a base. Any 17#-hydroxy group in the products may be esterified, or, where appropriate, oxidized to OXO. O-(f-nitrophenyl)hydroxylamine is prepared by the action of trifluoroacetic acid on tert-butyl N-(f-nitrophenoxy)carbomate which itself is prepared by reaction of tert-butyl N-hydroxycarbamate with f-fluoronitrobenzene. Compounds I are said to possess anabolic, androgenic and antilittering properties, and may be made up with carriers in to pharmaceutical compositions for oral administration.
