abstract 1297386 Dibenzothiepin and dibenzoxepin derivatives CIBA GEIGY AG 26 Nov 1969 [27 Nov 1968] 57849/69 Heading C2C Novel compounds of Formula I: (in which X is O or S, Y is H, Cl, CH 3 , CH 3 O or CH 3 S and R is H, straight chain C 1-6 alkyl, isopropyl or allyl) and their pharmaceutically acceptable acid addition salts, are prepared (i) when R is not H, by reacting a compound of the Formula II: with an amine of formula RNH 2 (in which R is as defined above except for H), or (ii) when R is H, by hydrolysing a compound of Formula III: (in which R is CN or the acyl radical of an organic acid), and, if desired, converting a compound I thus obtained into a pharmaceutically acceptable acid addition salt. Starting materials of Formula III in which Y is ethoxycarbonyl, are prepared by reacting a compound of Formula IV; (in which R 2 is C 1-6 alkyl, allyl or benzyl) with ethyl chloroformate. Pharmaceutical compositions having central depressant and antihistaminic activity, for oral, rectal or parenteral administration, comprise a compound of Formula I, or a pharmaceutically acceptable acid addition salt thereof, together with a pharmaceutically acceptable diluent or carrier.
