titleOfInvention | Process for the preparation of nucleoside-5'-diphosphates and their esters |
abstract | 1285312 Nucleoside-5<SP>1</SP>-diphosphates BOEHRINGER MANNHEIM GmbH 19 April 1971 [3 April 1970] 26008/71 Heading C2P Nucleoside- 5<SP>1</SP>-phosphates and esters thereof are prepared by reacting a free acid or salt, the anion of which has the formula where X is a nucleoside residue and Am is -NR 1 R 2 where R 1 and R 2 are hydrogen or C 1-4 alkyl or where R 1 and R 2 form a heterocyclic ring which may contain a further heteroatom, with a free acid or salt, the anion of which has the formula O 3 P<SP>2-</SP>-O-R (II) where R is hydrogen or the residue of a hydroxyl groupcontaining organic compound, the reaction being carried out in a C 1-4 aliphatic alcohol and/or in a di(C 1-3 alkyl) ketone and/or in dimethyl formamide, the solvent or solvent mixture optionally containing water, at a pH of 1 to 6À5. Specified acids having anions of Formula I are cytidine-5<SP>1</SP>-phosphoric acid morpholidate, adenosine-5<SP>1</SP>-phosphoric acid morpholidate and uridine-5<SP>1</SP>-phosphoric acid piperidate. Specified (phosphoric acid esters are chloine phosphate, 2-aminoethyl phosphate, serine phosphate, glucose-1-phosphate, glucose-6-phosphate, galactose-1-phosphate mannose-1-phosphate, glucosamine phosphate and N-acetyl glucosamine phosphate, and the calcium and mixed calcium chloride salts of these. The nucleoside diphosphates may be neutralized to form salts, for example sodium salts. |
