A reagent for determination of alkaline phosphatase in serum comprises an aqueous solution of para-nitrophenyl phosphate containing diethanolamine hydrochloride in a molar concentration of at least 1.5 and at most 4.5. In an Example the reagent also contains sodium azide and magnesium chloride and is adjusted to a pH of 9.8. In carrying out a test the serum sample and the reagent are mixed and the rate of change of extinction of the mixture is measured at 405 nm.
