abstract | 1,266,566. Bicyclooctane-carboxylic acid derivatives. E. I. DU PONT DE NEMOURS & C O . 23 April, 1969 [23 April, 1968; 24 Feb., 1969], No. 20855/69. -Heading C2C. Novel compounds I wherein two of the A's are single bonds and the other is a single or double bond or if Y is COOalkyl two of the A's are double bonds and the other is a single bond; R 1 -R 4 are H, F, Cl, Br, 1-12 C alkyl, provided (a) that R 1 -R 4 together contain not more than 16 C (b) at least one of R 1 -R 4 is alkyl and (c) not more than two of R 1 -R 4 are halo and that when two are halo they are adjacent to the same A; X is H, OH, OCOR<SP>6</SP>, (R 6 is H or 1-12 C alkyl), 0R 5 (R 5 is 1-6 C alkyl), 3-6 C cycloalkyl, 2-tetrahydropyrenyl or -CH 2 OCH 3 ; Y is -CHO, a group hydrolysable to -CHO, -COOR 7 (R 7 is H, 1-12 C alkyl, tetrahydropyranyl or a cation) -CH 2 OR 8 (R8 is H, hydrolysable esterifying group or an ether forming group) or COR 9 (R 9 is NHOH, NR 10 R 11 or NHNR 10 R 11 ; R 10 and R 11 are H, CH 3 , C 2 H 5 or C 3 H 7 or NR 10 R 11 is pyrrolidino, piperidino or morpholino) and R 12 is H, F, Cl, Br or CH 3 are prepared by (a) reducing a suitable compound I wherein one of R 1 - R 4 is an acyl group to give I where all A's are single bonds and at least one of R 1 to R 4 is a 2-12 C alkyl group; (b) reacting a compound II with a compound III wherein R 7 is 1-12 C alkyl and A is a single or double bond followed where necessary by reduction of compounds I where more than one A is a double bond; and (c) converting any group X or Y into another group X or Y by methods known per se. Intermediates isolated are compounds IV which are prepared by reaction of an appropriate alkyl phenyl ketone and an alkoxymeth. yiene-malonic acid diester; compounds II which are prepared by reaction of compounds IV with compounds R 3 -CH = CH-R 4 and the compounds used as starting materials in process (a) above which are prepared in a similar manner to compounds II. Pharmaceutical compositions useful in the prevention of pregnancy comprise a compound I together with a suitable carrier for oral administration. |
