abstract | 1,266,255. Dried agarose gel. MARINE COLLOIDS, Inc. 17 March, 1970, No. 12727/70. Heading C3U. The capacity of a dried agarose gel matrix consisting essentially of agarose whose cell structure is that determined by formation from an aqueous solution containing from 0À5 to 5% of agarose to form, when rehydrated, an aqueous gel wherein the percentage by weight of agarose corresponds to that of the original gel (within 20%), is improved by including in the gel structure at least 0À1% by weight of a macromolecular, predominantly organic hydrocolloid occluded and dried in situ. The hydrocolloid is required to be water-soluble to the extent of at least a 1% solution at a temperature below that at which the agarose has any appreciable solubility, so that the hydrocolloid is removable by aqueous leaching when the gel is rehydrated. The hydrocolloid, which may have a molecular weight between 5000 and 1 x 10<SP>6</SP>, is exemplified by amylopectin, sodium alginate, potassium #- carragheenate, carragheenan, deacetylated chitin, okra mucilage, guar gum, gum tragacanth, carboxymethyl locust bean gum, pectinic acid amide, (poly)D-mannose (phosphated), (poly)- D-glucose, D-galactose and D-mannuronic acid (acetylated), (poly)D-mannose, D-xylose and D-glucuronic acid (acetylated), hydroxyethyl cellulose, sodium carboxymethylcellulose, polyethylene oxide and polyvinyl pyrrolidone. The gel may be dried with the aid of a volatile hydrophilic liquid such as acetone, methyl ethyl ketone, methanol, ethanol, propanol and isopropanol. The dried gel may be in the form of fine particles, or a layer or film. |
