The invention comprises a 4-unsubstituted rifamycin of formula wherein X is (a) -CH 2 NB or -CH=NB in which NB is a substituted or unsubstituted amino group or is pyrrolidino, piperidino or morpholino or (b) -CH=NR in which R is a 4-methyl piperazino radical or a substituted or unsubstituted amino group; and R<SP>1</SP> is H or, when X is -CH=#+NB, a negative charge. The compounds, wherein X is -CH 2 NB are prepared by treating 4-desoxy-4-diazo-rifamycin 5 with formaldehyde and an amine of formula HNB wherein NB has the above meaning. Subsequent treatment with (a) manganese dioxide provides compounds wherein X is -CH=+#NB and then (b) a hydrazine H 2 NR in which R is as defined in claim 1 provides compounds wherein X is CH=NR.
