abstract | 1,253,339. Prothrombin complex. BAXTER LABORATORIES Inc. 28 May, 1970 [19 June, 1969], No. 25674/70. Heading A5B. A prothrombin complex useful in the treatment of haemorrhagic diseases is prepared by suspending a Cohn Plasma Fraction II+III, III, III-O, IV-1 or IV-1+IV-4 in physiological saline, adjusting the pH to 6.8-7.2, adding to the supernatant tribasic calcium phosphate, collecting the precipitate (containing adsorbed protein) and mixing it with 0.05M to 0.2M trisodium citrate solution, collecting the supernatant and subjecting it to two successive precipitations with polyethylene glycol of M.W. from 200 to 20,000, first at pH 6.8-8.0 and a final concentration of 5-10% PEG (precipitate rejected) and then at pH 5.0-5.4 and a final PEG concentration of at least 20%, the second precipitate, which is the complex, containing the prothrombin being collected. The final complex may be suspended in citrated saline for direct use by injection or, lyophilized to give a storable product. |
