titleOfInvention New basically substituted coumarin derivatives and process for the production thereof
abstract 1,206,572. Coumarin derivatives. CASSELLA FARBWERKE MAINKUR A.G. 17 Jan., 1969 [19 Jan., 1968], No. 2868/69. Heading C2C. Novel coumarin derivatives of the formula and pharmaceutically acceptable salts thereof, wherein R is the radical of an aliphatic, cycloaliphatic, araliphatic or aromatic amine or of a 5- or 6-membered heterocyclic nitrogen base free of acyloxy groups, which is bound via a nitrogen atom, R 1 is alkyl or aryl, R 2 is 5,7-, 6,7- or 7,8-positioned alkoxy groups, R 3 is alkoxy, and m is 1, 2 or 3, are obtained (a) by acylating, if desired in the presence of an acidbinding agent, compounds of the formula where R<SP>1</SP> is the same as R above except that the heterocyclic base is free from OH groups, with an alkoxybenzoic acid HOOC.Ph.(R 3 ) m (where Ph is the benzene nucleus) or with a functional derivative thereof, and eliminating by hydrogenation or saponification any protective groups in R<SP>1</SP>; or (b) by reacting, optionally in the presence of an acid-binding agent, a compound of formula (where Hal represents a halogen atom) with an amine of the formula RH. Coumarin derivatives of Formula (II) are obtained by reacting the corresponding amines with the 3-γ-halo-#-hydroxypropyl-coumarins, or with the corresponding 3-(2<SP>1</SP>,3<SP>1</SP>-epoxypropyl)- coumarins. Coumarin derivatives of Formula (IV) are obtained by acylating the corresponding 3-γ- halogen - # - hydroxypropyl - coumarins with an alkoxybenzoic acid HOOC.Ph(R 3 ) m or a functional derivative thereof. Pharmaceutical compositions having activity as coronary dilators comprise a compound of the Formula (I) or a pharmaceutically acceptable salt thereof, together with conventional pharmaceutical carriers, in forms suitable for oral or parenteral administration.
