abstract | 1,188,985. Analytical test pack. E.I. DU PONT DE NEMOURS & CO. 18 April, 1967 [26 April, 1966], No. 17677/67. Heading B1X. An analytical test pack comprises a container formed from pliable polymeric material, the container being divided into one or more closed compartments 10 (Fig. 1) or 24 (Fig. 4) and a reaction chamber, means communicating with the reaction chamber for introducing the test sample, the compartments 10 or 24 being independently openable into communication with the reaction chamber, and at least one compartment containing an analytical reagent. In the embodiment of Figure 1, compartments 10 are formed as depressions in one of two sheets of polymeric material used to make the container, the sheets being bonded together in the areas surrounding each depression and around their peripheries, the bonded areas between the compartments and the reaction chamber 28 being rupturable by manual pressure. The means for introducing the test sample comprises rigid header 13, having cylindrical channel 14 through it and opening (15) to its outside, the header being bonded to one of the sheets of polymeric material. Cartridge 18 inserted into channel 14 has passage 21 which communicates via hole (15) in header 13 and an opening in the adjacent sheet of polymeric material with the reaction chamber. The test sample may be injected through hole 16a in header 13 and through stopper 20 into channel 21. Alternatively the sample may be injected through hole 16b into the main part of tube 18 and there treated, for example with ion exchange resin and/or by filtration through a partition 23 before flowing into the reaction chamber. In the embodiment of Figure 4, clips 25 are used to close the reaction compartments, and the means for introducing the test sample comprises flexible tube 22 sealed between the two sheets of polymeric material. Examples relate to determining urea in blood serum using 1-cysteine hydrochloride, sodium salts of alpha-ketoglutaric acid and of dihydrobetadiphosphopyridine nucleotide, alpha-glutamic dehydrogenate and urease; determination of dehydrogenases (lactic acid, malic acid and alpha-hydroxybutyric acid dehydrogenases) using the corresponding acids, beta-nicotinamide adenine dinucleotide, phenazine methosulphate and 2, 6-dichlorophenol indophenol; determining glucose in e.g. blood serum using glucose oxidase, peroxidase and 3, 3<SP>1</SP> dimethoxybenzidine hydrochloride; determining glutamic-oxalacetic or glutamic-pyruvic transaminases in blood serum using beta nicotinamide adenine dinucleotide, phenazine methosulphate, 2, b-dichlorophenol indophenol, 1-glutamic dehydrogenase, alpha-keto glutaric acid, 1-aspartic acid and 1-alanine. For the latter determination, cartridge 18 is loaded with polyacrylamide gel and the sample injected into this and washed through into the reaction chamber with phosphate buffer.
