abstract | 1,179,435. Determination of rheumatoid factor. ORTHO PHARMACEUTICAL CORP. 18 May, 1967 [17 June, 1966], No. 23157/67. Heading B1X. A reagent for determining rheumatoid factor comprises a suspension in a polar liquid of solid synthetic polymer particles having average diameter 0.15 to 14 microns and coated with rabbit gamma-globulin which has been subjected to the action of proteolytic enzymes. The polymer particles are preferably polystyrene particles average diameter 0.81 micron; acrylic polymer is mentioned. The enzyme is preferably trypsin, chymotrypsin or papain; also mentioned are fungal proteases and pepsin. In preparing the reagent, the rabbit gamma-globulin in phosphate or bromate buffer pH 8.6 is mixed with the proteolytic enzyme then the treated gamma-globulin is added to the particle suspension. The suspension may then be heated, for example to 50C for 7 to 10 days to complete the treatment of the gamma-globulin. |
